AbstractAsthma phenotypes have been developed to address the complexities of the disease. However, owing to a lack of longitudinal studies, little is known about the onset as well as the stability of phenotypes. Distinguishing phenotypes with regard to the severity or duration of the disease is essential. A phenotype covers the clinically relevant properties of the disease, but does not show the direct relationship to disease etiology and pathophysiology. Conclusions: A discrepancy between subjective assessment of asthma control and ACT score was observed in the examined group of patients. The frequency of asthma exacerbations (according to questionnaire) was describing the degree of asthma control more precisely than self-assessment. Older age and longer disease duration were combined with significantly worse asthma control. Better asthma control was combined with the presence of influenza vaccinations in medical history.

Lipopolysaccharide enhances FcɛRI‐mediated mast cell degranulation by increasing Ca2+ entry through store‐operated Ca2+ channels: implications for LPS exacerbating allergic asthma

This study investigated the effect of LPS on antigen‐stimulated mast cell degranulation and the underlying mechanisms. We found that LPS enhanced degranulation in RBL‐2H3 cells and mouse peritoneal mast cells upon FcɛRI activation, in dose and time‐dependent manners. Parallel to the alteration of degranulation, LPS increased FcɛRI‐activated Ca2+ mobilization, as well as Ca2+ entry through store‐operated calcium channel (SOC) evoked by thapsigargin (TG). Blocking Ca2+ entry through SOCs completely abolished LPS enhancement of mast cell degranulation. Consistent with functional alteration of SOCs, LPS time‐dependently increased mRNA and protein levels of Orai1 and STIM1, two major subunits of SOCs. In conclusion, salbutamol can decrease acute and chronic inflammation, possibly through the stimulation of β-2 adrenergic receptors. This anti-inflammatory effect may be of significance in asthma treatment, where inflammation also takes part in the etiopathology.
